潞党参多糖对宫颈癌细胞SiHa增殖和迁移的影响

Effects of Polysaccharides from Lu Dangshen(Codonopsis pilosula)on Proliferation and Migration of Human Cervical SiHa

摘要: 以上党地区道地药材潞党参的多糖成分(Lu Dangshen polysaccharides,LDP)为对象,研究其对人宫颈癌细胞SiHa的增殖及迁移的影响。通过MTT法检测LDP对SiHa细胞增殖的影响。通过细胞黏附试验、铺展试验和划痕试验,分析LDP对SiHa细胞迁移的影响。结果显示,LDP显著抑制了SiHa细胞的增殖,IC50值为0.68 mg/mL;LDP明显抑制了SiHa细胞与外基质的黏附作用,延缓了其铺展过程,并抑制了其体外迁移运动能力。LDP对人宫颈癌细胞SiHa的增殖、黏附、铺展以及迁移运动能力具有显著的抑制效应,具有开发为肿瘤治疗的辅助药物及保健食品的价值。

Abstract: The effects of polysaccharides from Lu Dangshen(Codonopsis pilosula)(LDP)on the proliferation and migration of human cervical cancer cell SiHa was investigated. MTT method was used to determine the effects of LDP on SiHa cell proliferation,and cell adhesion assay,cell spreading assay and cell wound scratch assay were used to analyze the effect of LDP on SiHa cell migration. LDP significantly repressed SiHa cell proliferation(IC50 = 0.68 mg/mL),and inhibited adhesion between cells and extracellular matrix,and delayed cell spreading,and then restrained SiHa cell migration. LDP presents solid inhibition effects on the proliferation,adhesion,spreading and migration of human cervical cancer cell SiHa,and thus possesses potential value for developing auxiliary medicine for tumor treatment and dietary supplement.
