Immunoregulatory Effect of Polysaccharides from Codonopsis pilosula on the Ana-1 Macrophages in Mice

Abstract: The immunoregulatory effect of polysaccharides from Codonopsis pilosula on the Ana-1 macrophages of mice was investigated. MTT method was used to determine the effects of C. pilosula polysaccharides on Ana-1 cell proliferation. ELISA assay was applied to detect the effects of C. pilosula polysaccharides at different doses on TNF-α and IL-1β secretion in Ana-1 macrophages. C. pilosula polysaccharides at different doses were given to mice by intrapefitoneal injection,then the thymus index,proliferation of lymphoma cells,and the levels of TNF-α and IL-1β in the serum of mice were determined. The in vitro test showed that C. pilosula polysaccharides significantly increased TNF-α and IL-1β secretion of Ana-1 macrophages in a dose-dependent way,but presented no effect on Ana-1 cell proliferation. The in vivo tests indicated that the spleen index,lymphocyte proliferation index,and the levels of TNF-α and IL-1β in the serum of middle-dose and high-dose polysaccharides groups significantly increased in a dose-dependent way,compared to control mice. Therefore,C. pilosula polysaccharides may effectively improve the immune activity of Ana-1 macrophages in mice,i.e.,improve the immune functions of mice.
