动力学模型在荧光定量PCR 数据处理中的优势

Advantages of the Dynamic Model in Real-time PCR Data Processing

摘要: 近年来荧光定量PCR 技术凭借其基因定量的优势而得到了广泛应用,上游技术的发展保障了原始数据的精确性,但定量结果的可靠性仍取决于PCR 扩增效率和所采用的数据处理模型,基于不同数学模型对原始数据处理后的结果会有较大差异。试验通过对样品进行5 倍梯度稀释后获得相应梯度的荧光定量PCR 原始数据,并应用2 -△△ Ct 法、相对标准曲线法和动力学模型处理原始数据,所得定量结果(x-±s)依次为1.76±0.70、0.89±0.14 和0.99975±0.06,与理论值1 比较表明,动力学模型在定量结果的可靠性、方便性及经费节约方面都具有良好应用前景。

Abstract: In recent years, Real-time PCR with its advantages in the quantitation of gene is widely used, the development of equipment ensure the accuracy of raw data, however the reliability of quantitative result highly depends on the PCR efficiency and the mathematical model on which the quantitative methods are based, The quantitative result processed with different mathematical models will be quite different. In this paper, the sample was 5-fold diluted for gaining the raw data of quantitative PCR, and the same raw data was processed by three kinds of mathematical models, including 2 -△△ Ct model, relative standard curve model, dynamic model. And the results demonstrate the dynamic model has a good prospect.
