党参多糖对Ana-1巨噬细胞和小鼠的免疫调节作用

Immunoregulatory Effect of Polysaccharides from Codonopsis pilosula on the Ana-1 Macrophages in Mice

摘要: 以中药党参为材料,研究其多糖成分对小鼠巨噬细胞Ana-1和小鼠的免疫活性的影响。通过MTT实验,检测党参多糖对Ana-1细胞增殖的影响。利用ELISA实验,检测不同浓度的党参多糖对Ana-1细胞释放TNF-α和IL-1β的影响。用不同剂量的党参多糖灌胃小鼠,分别测定小鼠脾脏指数、淋巴细胞增殖活力以及血清中TNF-α和IL-1β的含量。体外实验结果显示党参多糖不影响Ana-1细胞的增殖,却显著提高TNF-α和IL-1β的分泌,并呈浓度依赖关系。体内实验结果显示:与对照组相比,党参多糖中、高剂量组小鼠胸腺指数、淋巴细胞增殖指数以及血清中TNF-α和IL-1β的含量均显著增高,呈剂量依赖关系。因此,党参多糖能够有效提高小鼠巨噬细胞Ana-1的免疫活力,增强小鼠的免疫功能。

Abstract: The immunoregulatory effect of polysaccharides from Codonopsis pilosula on the Ana-1 macrophages of mice was investigated. MTT method was used to determine the effects of C. pilosula polysaccharides on Ana-1 cell proliferation. ELISA assay was applied to detect the effects of C. pilosula polysaccharides at different doses on TNF-α and IL-1β secretion in Ana-1 macrophages. C. pilosula polysaccharides at different doses were given to mice by intrapefitoneal injection,then the thymus index,proliferation of lymphoma cells,and the levels of TNF-α and IL-1β in the serum of mice were determined. The in vitro test showed that C. pilosula polysaccharides significantly increased TNF-α and IL-1β secretion of Ana-1 macrophages in a dose-dependent way,but presented no effect on Ana-1 cell proliferation. The in vivo tests indicated that the spleen index,lymphocyte proliferation index,and the levels of TNF-α and IL-1β in the serum of middle-dose and high-dose polysaccharides groups significantly increased in a dose-dependent way,compared to control mice. Therefore,C. pilosula polysaccharides may effectively improve the immune activity of Ana-1 macrophages in mice,i.e.,improve the immune functions of mice.
