Comparison of Biological Characteristics of Feline Mesenchymal Stem Cells Derived from Four Different Tissues

Abstract: To compare the biological characteristics of Mesenchymal Stem Cells(MSC)derived from adipose,bone marrow,amniotic membrane and umbilical cord,whole blood culture was used to isolate bone marrow-derived MSC and digestion to isolate others-derived MSCs. Then the morphology,growth characteristics,differentiation capacity,and cell surface markers were compared and analyzed. The results showed that feline MSC derived from 4 different tissues had common characteristics of adherent growth,spindle-shaped and polygonal-shaped. They all had the ability of adipogenic and osteogenic differentiation. The cell proliferation rate of adipose derived-MSC and bone marrow derived-MSC were faster than amniotic membrane derived-MSC and umbilical cord derived-MSC,and the doubling time of MSC from adipose and bone marrow was shorter. The adipose derived-MSC demonstrated the best growth activity and potential application prospect at clinic,because it maintained fine proliferative activity when they passed to the 9th generation. Cell surface markers of CD44,CD90 and CD105 were higher expressed,and CD34 were less expressed in four kinds of MSC.
