Preliminary Study on Inducing Axillary Buds of Different Pinus serotina Explants

Abstract: The varied segments from field-growing 1-year old Pinus serotina spring stem and shoot tips from seed germination were used as explants. The explants were disinfected by 0.1% HgCl2,and those with favorable disinfection were used as sterile materials,and then the combination of medium type and the hormones for the axillary bud induction were optimized. The results showed that 10 min disinfection of middle parts of the spring stems by 0.1% HgCl2 was the optimal,the pollution rate was the lowest(0.8),and the best combination of medium and hormone levels to induce axillary buds was MS medium with 2.5 mg/L 6-BA and 0.2 mg/L NAA. And the optimal disinfected time of seed was 4 min,the pollution rate was 2.4%,and the induction rate was the highest in the medium containing 0.7 mg/L NAA and 1.0 mg/L 6-BA. In summary,an effective method to induce the axillary buds from the spring stem and seeds is established,which may provide theoretical evidences and technical support for the rapid and efficient breeding of Pinus serotina.
