堆肥过程中纤维素降解菌的分离与鉴定

Isolation and Identification of Cellulose Degrading Microorganisms in Composting Process

摘要: 为了解决好氧堆肥中秸秆降解缓慢的问题,从玉米秸秆堆肥中筛选高效降解纤维素的菌株。采用平板稀释法并结合刚果红染色法对玉米秸秆堆肥中的纤维素降解菌进行初筛,通过滤纸条崩解实验进行复筛,筛得滤纸崩解能力较高的2株放线菌C31和C37,经鉴定分别为放线菌(Streptomyces drozdowiczii)和黄麻链霉菌(Streptomyces corchorusii);1株细菌GD16,经鉴定为类芽孢杆菌(Paenibacillus pabuli)。对3株菌株进行了酶活的定量测量,其中菌株C31具有的纤维素酶活性和滤纸酶活性最高,分别为4.8 U/mL、3 U/mL,菌株GD16具有的木聚糖酶活性最高可达到23 U/mL。结果显示菌株C31、C37和GD16均具有纤维素酶活性、滤纸酶活性和木聚糖酶活性。

Abstract: In order to solve the problem of slow degradation of straw in aerobic compost,strains that efficiently degrade cellulose were screened from corn straw compost. The cellulolytic bacteria in the corn stover compost were screened by the plate dilution method combined with the Congo red staining method,and the re-screening was conducted by the filter paper disintegration test. Two actinomycetes C31 and C37 with high filter paper disintegration ability were screened and identified as Streptomyces drozdowiczii and Streptomyces corchorusii. Strain GD16 was identified as Paenibacillus pabuli. At the same time,the enzyme activities of three strains were quantitatively measured. The strain C31 had the highest cellulase activity and filter paper enzyme activity,which were 4.8 U/mL and 3 U/mL,respectively. The strain GD16 had the highest xylanase activity,and reached 23 U/mL. The results showed that strain C31,C37 and GD16 all had cellulase activity,filter paper enzyme activity and xylanase activity.
