Optimum Technique of Extracting Mycelium Polysaccharide from Auricularia auricular Using Box-Behnken Design-Response Surface Methodology

Abstract: To optimize the extraction technique for mycelium polysaccharide from A. auricular, the effects of extraction temperature, extraction time, the liquid to solid ratio and their interaction on extraction rate were studied by Box-Behnken design. The predictive model and reliability were developed by Design Expert software and response surface analysis(RSM). The optimal extraction conditions were achieved and listed as follows: extraction temperature 81.59℃, extraction time 81.40 min, liquid-to-solid ratio 36.42:. Under the optimum conditions, the extraction yield of mycelium polysaccharide from A. auricular reached 7.79 mg/g, while the predicted one was 7.92 mg/g. The results showed the suitability of RSM in optimizing the extraction of mycelium polysaccharide from A. auricular. The necessary technical support was provided by this experimental study for the industrial production of mycelium polysaccharide from A. auricular.
