Evaluating the Development of Small Interfering RNA Expression Vector from Its Functional Elements

Abstract: RNA interference as a potent gene silencing approach plays important parts in gene regulation, functional study and gene therapy. The small interfering RNA expression vector achieves a sustainable, stable and controllable application of the RNAi technology, and becomes a promising way for gene silencing. Although the interfering RNA expression vector has progressed to the second generation, and many commercial products were availabe, the discrepancies among efficiency, safety and controllability of these vectors still exist. The development of the interfering RNA expression vector seems get into a lag phase. Therefore, this article analysed the history and development of these small interference RNA expression vectors on the basis of their functional blocks, providing a theoretical fundation for the vector’s optimization and selection.
