榆黄菇遗传多样性的ISSR和SRAP综合分析

摘要: 榆黄菇味道鲜美,营养丰富,且必需氨基酸含量高,近几年成为食用菌的重要栽培种类之一。通过ISSR和SRAP分子标记方法对全国收集到的19个榆黄菇品种进行遗传多样性综合分析,从而为其杂交育种的亲本选择提供理论指导。筛选出10个ISSR引物和10对SRAP引物,利用筛选的引物,以榆黄菇的基因组DNA为模版进行PCR扩增,共获得194条清晰条带,其中150条是多态性条带,占总条带的77.3%。对条带进行聚类分析,结果表明在相似系数为0.70水平上,可将19个供试榆黄菇菌株分为4大类,表明19个供试榆黄菇菌株间具有较大的遗传差异,聚类分析图为杂交育种的亲本选择提供分子水平的依据。

Abstract: The Pleurotus citrinopileatus have been one of the most important cultivated edible mushrooms in recent years,because they taste delicious,are full of nutriments,and contain high essential amino acids. Genetic diversity of 19 P. citrinopileatus cultivars,collected from all over the China,was analyzed using combined ISSR and SRAP molecular markers in the experiment,which provides theoretical guidance for the parental selection of cross breeding. Ten ISSR primers and 10 SRAP primer pairs were selected for the producing reproductive bands using genomic DNA of P. citrinopileatus as templates. In total,194 amplification patterns were obtained,and in which 150 bands were polymorphic specific,accounting for 77.3% of all bands. The obtained results by cluster analysis indicated that the tested strains fell into four clades at a coefficient of 0.70,revealing that there was significant genetic diversity among P. citrinopileatus cultivars. The diagram of cluster analysis laid the foundation for the parental selection in the cross breeding of P. citrinopileatus.
