The Isolation and Identification of an Efficient Fe/Mn-oxidizing Bacterial Strain P1,and the Optimization of Its Oxidizing Conditions

Abstract: By enrichment culture,a Fe/Mn-oxidizing bacterial strain P1 was isolated from the sludge samples of groundwater well that was rich in Fe/Mn. According to morphologic and physiological-biochemical characteristics as well as 16S rDNA sequence analysis,strain P1 was identified as Bacillus cereus. Concurrently,single factor test was used to study the growth of strain P1 and its oxidation characteristics;and the response surface methodology(RSM)was employed to explore the effects of inoculation size,temperature and pH on the oxidation characteristics of strain P1 and further optimize the oxidation conditions. The results showed that the optimal oxidation conditions were temperature 28.54℃,pH 7.23,and inoculation size 4.35 %. At the optimal conditions,the removal ratios of Mn and Fe were 93 % and 100 % respectively in the selective medium containing 200 mg/L Fe and 800 mg/L Mn after strain P1 was cultured for 3 d.
