Isolation and Biological Characterization of Duck Adipose-derived Mesenchymal Stem Cells

Abstract: This study aims to establish in vitro culture system for the poultry adipose-derived stem cells(ADSCs),to study the biological characteristics and the multi-differentiation potential. Enzyme digestion method was applied to separate Beijing duck 18-day-old embryos ADSCs and to draw the growth curve. ADSCs were identified by RT-PCR,immunofluorescence and flow cytometric analysis. Our results suggested that the ADSCs had solid proliferative activity and were positive for CD29,CD44 and CD105,the rate of CD29+,CD44+,and CD105+ ADSCs was 95.39%,94.53%,98.19%,respectively. ADSCs differentiated into adipocytes and osteoblasts by induced differentiation in vitro. In summary,under appropriate experimental circumstances,Beijing duck embryo ADSCs have a strong self-renewing ability and multi-differentiation potential in vitro,and it can be an ideal type of seed cell for cellular transplantation therapy.
