摘要: 以常见的几种热带水果(香蕉、番木瓜、芒果、菠萝和荔枝等)为例,总结了近年来利用常见的技术手段如cDNA末端快速克隆技术(repid-amplification of cDNA ends,RACE)和实时逆转录聚合酶链式反应技术(reverse transcription-polymerase chain reaction,RT-PCR)等方式进行基因分离,并将成功克隆出的目的基因或片段进行基因分析与表达研究,最后阐述了基因分离与克隆技术的优势与缺陷,对基因分离及克隆技术提出了展望,旨为今后科研人员进行进一步的基因分子生物学水平上的热带果树研究与热带农业科学研究奠定基础。

Abstract: Firstly, we summarized the isolations of the genes from several common tropical fruits' genes(banana, papaya, mango, pineapple, litchi, etc.)by frequently used technologies of rapid-amplification of cDNA ends(RACE)and reverse transcription-polymerase chain reaction(RT-PCR), also the analyses and expressions of cloned target genes and fragments. Finally, we discussed the advantages and disadvantages of gene isolating and cloning technologies, and prospected the new gene isolating and cloning technologies, aiming at laying the foundation for studying tropical fruit tree and agriculture at biological level of genetic molecule.
