Research Progress of Bacillus subtilis Expression System and Its Promoter Regulatory Elements

Abstract: As a Gram-positive bacteria, Bacillus subtilis is an attractive host for the production of heterologous secretory proteins for several reasons:it is non-pathogenic and the capable of secreting functional extracellular proteins directly to the culture medium, a great deal of vital information concerning large scale fermentation and production technology. One of the key factors for achieving high-level expression of heterologous proteins is the use of a strong and control promoter. In present, the promoters of Bacillus subtilis can be classified into three categories:constitutive promoters, inducer-specific promoters and autoinducible promoters. This paper described the advantages and disadvantages of Bacillus subtilis expression system and the classification of promoters. At the same time, we summarized the methods of the amplification of new promoters, which provided a foundation for improving Bacillus subtilis expression systems and the industrial production of heterologous proteins .
