Evaluation of the Genetic Diversity of Pleurotus citrinopileatus by Combined ISSR and SRAP Analyses

Abstract: The Pleurotus citrinopileatus have been one of the most important cultivated edible mushrooms in recent years,because they taste delicious,are full of nutriments,and contain high essential amino acids. Genetic diversity of 19 P. citrinopileatus cultivars,collected from all over the China,was analyzed using combined ISSR and SRAP molecular markers in the experiment,which provides theoretical guidance for the parental selection of cross breeding. Ten ISSR primers and 10 SRAP primer pairs were selected for the producing reproductive bands using genomic DNA of P. citrinopileatus as templates. In total,194 amplification patterns were obtained,and in which 150 bands were polymorphic specific,accounting for 77.3% of all bands. The obtained results by cluster analysis indicated that the tested strains fell into four clades at a coefficient of 0.70,revealing that there was significant genetic diversity among P. citrinopileatus cultivars. The diagram of cluster analysis laid the foundation for the parental selection in the cross breeding of P. citrinopileatus.
